Penelitian ini bertujuan untuk mengetahui nilai total fenol dan flavonoid serta aktivitas antioksidan dengan menggunakan metode 1,1-difenil-2-pikrilhidrazil (DPPH) dari limbah cair tahu yang telah difermentasi menggunakan beberapa mikroba, yaitu Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5 dan Paecylomyces marquandii strain TP4, dengan molase sebagai nutrisi tambahan dan dikeringkan menggunakan alat freeze dry. Hasil uji fitokimia seluruh produk fermentasi menunjukkan negatif mengandung senyawa alkaloid, positif mengandung senyawa flavonoid dan saponin, serta positif mengandung senyawa fenol kecuali pada isolat Nocardia niigatensis strain TP5, dan positif mengandung senyawa tannin pada campuran Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5 dengan Paecylomyces marquandii strain TP4. Hasil uji total fenol pada isolat Streptomyces malaysiensis strain TT41; Nocardia niigatensis strain TP5; Paecylomyces marquandii strain TP4 dan campuran Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5 dengan Paecylomyces marquandii strain TP4 secara berturut-turut sebesar 1,4947; 1,4576; 1,1710; 1,1672 mg/g GAE dan total flavonoid sebesar 0,5383; 1,4092; 0,4839; 0,6208 mg/g QE. Pada pengujian aktivitas antioksidan menggunakan metode DPPH (1,1-difenil-2-pikrilhidrazil) sampel dibuat beberapa varian konsentrasi (20, 40, 60, 100, 200, 500 ppm) dilarutkan menggunakan pelarut etanol, kemudian ditambahkan 1 mL DPPH 1 mM dan disimpan dalam ruang gelap selama 30 menit, lalu dilakukan pengukuran menggunakan spektrofotometer UV-Vis dengan panjang gelombang maksimum (515-520 nm). Hasil uji aktivitas antioksidan yang didapat dari perhitungan dan perbandingan dengan IC50 menunjukkan campuran Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5 dengan Paecylomyces marquandii strain TP4 memiliki aktivitas antioksidan yang paling tinggi.

This study aims to determine the total value of phenols and flavonoids and antioxidant activity using the 1,1-diphenyl-2-picrylhydrazyl method (DPPH) from tofu liquid waste that has been fermented using several microbes, that is Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5, and Paecylomyces marquandii strain TP4, with molasses as additional nutrients and dried using a freeze-dry tool. The phytochemical test results of fermented products were negative for alkaloids, positive for flavonoids and saponins compounds, and positive for phenolic compounds, except for the Nocardia niigatensis isolate strain TP5, and positive for tannin compounds in the mixture of Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5 with Paecylomyces marquandii strain TP4. Total phenol test results in the mixture of Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5 with Paecylomyces marquandii strain TP4 were 1,4947; 1,4576; 1,1710; 1,1672 mg/g GAE and total flavonoids of 0,5383; 1,4092; 0,4839; 0,6208 mg/g QE. In testing the antioxidant activity using the DPPH method (1,1-diphenyl-2-picrylhydrazyl), samples were made of several concentration variants (20,40,60,100,200,500 ppm) dissolved using ethanol solvent. Then added 1 mL DPPH 1 mM and stored in a dark room for 30 minutes. Then measurements were made using a UV-Vis spectrophotometer with a maximum wavelength (515-520 nm). The results of the antioxidant activity test obtained from calculations and comparisons with IC50 showed that the mixture of Streptomyces malaysiensis strain TT41, Nocardia niigatensis strain TP5 with Paecylomyces marquandii strain TP4 has highest antioxidant activity.
